Paramagnetic contrast media are the great target of study, but iodine contrast media presents, nowadays, a great use in clinical practice, keeping CIN (Contrast Induced Nephropathy) as a problem. Recently contrast media developments point to Theranostic and Multimodal Contrast Media as great potential in healthcare improvement. On the other hand, porphyrinic derivatives are, at this time, a great field of study, beyond in environmental applications, in medical applications, namely in PDT (Photodynamic Therapy) and MRI (Magnetic Resonance Imagaging). The aim of this PhD research plan is to develop a non-toxic contrast agent based on porphyrin derivatives with theranostic and multimodal potential. To reach the main objective we propose to: synthetized and full characterize iodinated-(metallo)porphyrinic derivatives; assess their X-ray beam attenuation ability; Assess Toxicity of the new compounds that shown good X-ray attenuation, based on cellular viability and proliferation, genotoxicity and DNA content; evaluate Photodynamic potential of the promising compounds; and finally, to assess in vivo biodistribution in mice, using CT scan systems as a non-invasive technique. This PhD work plan is a three years collaborative research project involving the Organic Chemistry, Natural Products and Food Stuffs (QOPNA) research unit in the Department of Chemistry, University of Aveiro (DQUA) and the unit of Metabolism, Nutrition and Endocrinology (MNE) of the Institute of Research and Innovation in Health (i3S), University of Porto.
